Job Summary

The Saba Lab at UCSF investigates the role of sphingolipids in health and disease, with particular interest in the pathophysiology and treatment of sphingosine phosphate lyase insufficiency syndrome (SPLIS). Specific goals include: (1) to determine the natural history of SPLIS; (2) generate murine models of disrupted sphingolipid metabolism; (3) generate human preclinical models of disrupted sphingolipid metabolism; (4) use mouse models to dissect the signaling pathways by which sphingolipids regulate homeostasis of organ function; (5) use human models such as neurally reprogrammed fibroblasts and induced pluripotent stem cells to reveal the role of sphingolipids in organ function; (7) use gene and cell therapies to rescue phenotypes associated with disrupted sphingolipid metabolism. This SRA 1 position is for an individual who will work on sphingolipid research in mouse models and cell lines. SRA duties will include: cell/tissue culture, biochemical assays (western blotting, immunoprecipitation, qPCR), DNA and RNA extraction and manipulation, PCR and agarose gel electrophoresis, generation of recombinant retroviruses/lentiviruses, adeno associated viruses, and transduction of fibroblasts, flow cytometry, maintenance of accurate/complete/legible records, organization and maintenance of mouse colonies (including computerized record-keeping, breeding, monitoring and genotyping), and experimental procedures using laboratory mice (blood sampling, intravenous/intraperitoneal injection, dissection, analysis of harvested tissues).
